I create VM with SKU E16-4s_v3 and when create the reservation there is no E16-4s_v3, the option is for v4 and v5.
Can i know if i purchase reservation E16-4s_v4 then this can be used for E16-4s_v3?

Unfortunately you can no longer purchase Reserved Instances (RIs) for Esv3 series, and E16-4s_v4 RI cannot be used for E16-4s_v3. Please see screenshot excerpt below from Transition guide for retired Azure Reserved VM Instances:
You may consider if using Savings Plan makes sense, or using newer VM size instead.

I have unused reservation for E16-4s v3 on region1, and the VM now located in region2 with same SKU.
Because the VM location in different region then the unused reservation can't be used. Also because the v3 can't be purchased then if the VM upgraded to E16-4s v4 then can i exchange the unused reservation (E16-4s v3 on region1) to E16-4s v4 on region2?
I think the saving plan is not suitable for me because the VM is always on 24hours every day.
Hi, you can exchange the unused E16-4s_v3 reservation for a E16-4s_v4 reservation in different region. So if you upgrade VM in region2 to E16-4s_v4 you can use new reservation.
When exchanging you are allowed to pick different region/VM size for the new purchase, please see screenshot excerpt below:
